Introduction

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been reported to play roles in many different cellular processes, such as cell adhesion, cell transformation, cell cycle checkpoint, and cell volume control. Knockout studies in mice suggest that this kinase may be a fundamental regulator of cardiac contractility and Ca(2+) handling in myocytes.

Description

Protein Kinase C alpha Human Recombinant produced in Sf9 is a glycosylated, polypeptide chain containing amino acids 2-672 and having a molecular mass of 76.7 KD. This protein is the full-length form of the protein with an amino terminal poly His-tag.
PKC-a is purified by proprietary chromatographic techniques.

Storage Buffer

PKC-a is supplied in 20mM HEPES pH 7.5, 250mM NaCl, 50% glycerol, 2mM EDTA, 2mM EGTA, 0.05%Triton X-100 and 5mM DTT.

Dilution Buffer

10mM HEPES (pH7.4), 0.01% CHAPS and 5mM DTT.
*for further dilution you may use either the Storage Buffer or Dilution Buffer.

Stability

Store at 4°C if entire vial will be used within 2-4 weeks.
Store, frozen at -20°C for longer periods of time.
Avoid multiple freeze-thaw cycles.

Purity

Greater than 95% as determined by SDS-PAGE.

Specific Activity

1570 nmole of phosphate transferred to Histone H1 substrate per minute per mg of total protein at 30°C. Activity determined at a final protein concentration of 0.5 to 2ug/ml.

Unit Definition

7200- 9000 units/mg. One unit of PKC-a activity transfers 1 nmole of phosphate to a PKC substrate peptide (myelin basic protein 4-14) per minute at 30°C. Recombinant human active PKC-a also phosphorylates full length myelin basic protein, histone, MARKS, and GSK-3b. Activity requires incubation with lipid activators, and may vary depending on the substrate and reaction conditions.
