Introduction

Nucleophosmin (NPM1) is a phosphoprotein which shuffles between the nucleus and the cytoplasm. NPM1 is assumed to be involved in a number of processes including regulation of the ARF/p53 pathway. Several genes fusion partners have been described, particularly the anaplastic lymphoma kinase gene on chromosome 2. NPM1 gene mutations are linked with acute myeloid leukemia.

Immunogen

Anti-human NPM1 mAb, is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with a recombinant human NPM1 protein 1-294 amino acids purified from E. coli.

Ig Subclass

Mouse IgG1 heavy chain and k light chain.

Purification Method

NPM1 antibody was purified from mouse ascitic fluids by protein-A affinity chromatography.

Formulation

1mg/ml containing PBS, pH-7.4, 10% Glycerol and 0.02% Sodium Azide.

Storage Procedures

For periods up to 1 month store at 4°C, for longer periods of time, store at -20°C. 
Prevent freeze thaw cycles.

Stability / Shelf Life

12 months at -20°C. 1 month at 4°C.

Applications

NPM1 antibody has been tested by ELISA, Western blot analysis, ICC/IF and Flow cytometry to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:1000. Recommended dilution range for ICC/IF and Flow cytometry is 1:200.
