Introduction
Caused by one of four closely related virus serotypes of the genus Flavivirus, family Flaviviridae, each serotype is sufficiently different that there is no cross-protection and epidemics caused by multiple serotypes (hyperendemicity) can occur. In cell culture experiments and mice Morpholino antisense oligos have shown specific activity against Dengue virus.
Description
Recombinant Dengue p18 antigen is a genetically engineered protein generated from dengue envelope protein, containing most dengue subtype epitopes. Specifically developed for dengue IgG/IgM lateral flow immunoassay. Recombinant Dengue p18 can detect dengue IgG and IgM from all four infected dengue virus subtypes. Having over 90% sensitivity and specify to detect dengue IgM and IgG. This antigen gives a good performance to detect dengue IgM with strong signal intensity.

Protein is >95% pure as determined by 10% PAGE (coomassie staining).
Formulation
Phosphate buffered saline, 0.02% sodium nitrate and 25mM K2CO3.
Stability
Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Avoid multiple freeze-thaw cycles.

Preparation protocol
Following is the protocol needed to develop the rapid test by this antigen:
1. Prepare λ522 to 525 fresh gold particle within 1-2 days, adjust OD to 1.2 to 1.4.
2. conjugate at pH 7.2-7.5 at a concentration of 16ug/ml;
3. conjugate for 2-4 hours, then add 25mM phosphate buffer with 0.2%PVP 10,000 pH8.0 to suspend the conjugate gold pellet.
4. Block the suspension with 10% BSA to final 1% BSA.
5. Use the sample pad treated with 50mM borate, 1% Tween-20 and 0.5% PVP10,000, pH9.2.
6. Prepare the sample buffer with 50-100 mM carbonate buffer with0.5-1.0% Tween-20.
The rapid test prepared by this antigen has over 90% sensitivity and specificity over 90% to detect dengue IgM and IgG.
