About CCL27:

CCL27 (Chemokine C-Cmotif ligand 27) or CTACK (cutaneous T-cell-attracting chemokine) is a cytokine in the CC chemokine family. It is also referred to as skinkine, ESkine and IL-11 R-alpha-locus chemokine (ILC). CTACK is related to the homing of memory T lymphocytes to the skin and helps with T cell-mediated inflammation in the skin. It can be found in various tissues, including skin, placenta and gonads. CTACK binds to the CCR10 chemokine receptor, and the gene is found on chromosome 9 in humans. CCL27 and CCR10 together are likely to play a role in skin homeostasis and possibly in inflammatory responses too. CCL27 is the ideal product for research into immune system processes and wound healing It is available in both human and mouse recombinants to enable more precise research.

CCL27 can be produced in a non-secreted form, which is unusual, and this is as a result of alternative splicing. The protein is called PESKY targets the nucleus and induces a rearrangement of the actin cytoskeleton, which enhances cell motility. Secreted CCL27 can also reach the nucleus after CCR10-mediated internalisation. This could modulate transcription to alter cell behaviour. CCL27 is produced by keratinocytes but can be induced by stimulation with TNF-alpha and IL-1beta. CCL27 can be detected in both healthy and infected skin.

CTACK has been observed in inflammatory skin conditions such as atopic dermatitis and psoriasis vulgaris. Serum CTACK levels in patients with these diseases have been measured as higher than in healthy control subjects. This suggests that CCL27 is an important chemokine for the pathogenesis of atopic dermatitis and psoriasis vulgaris. CCL27 is continually expressed by keratinocytes and as well as being in higher levels when atopic dermatitis and psoriasis are present, it is also elevated in patients who have onchocerciasis, an infection caused by a parasitic worm, which is also called river blindness. Higher levels of CCL27 are also detected in cases of allergic contact dermatitis.

In patients with alopecia areata, CCL27 immunopositivity has been shown to be lower than in normal scalp tissue. Topical immunotherapy, the most effective treatment currently known for this type of alopecia, demonstrates an increase in CCL27 expression. ILC/CCL27 could collectively offer a good candidate molecule for cancer gene therapy. CCL27 is also induced by UVB irradiation, from epidermal to dermal compartments. Increased BMDK migration cased by CTACK accelerates the healing of wounds without influencing angiogenesis or keratinocyte proliferation.
