Introduction
Nicastrin is a Type I transmembrane glycoprotein which is an integral component of the multimeric gamma-secretase complex. Nicastrin cleaves integral membrane proteins, including Notch receptors and beta-amyloid precursor protein, and may be a stabilizing cofactor required for gamma-secretase complex assembly. The cleavage of beta-amyloid precursor protein yields amyloid beta peptide which is the main component of the neuritic plaque and the hallmark lesion in the brains of patients with Alzheimer''s disease; however, the nature of the Nicastrin''s role in Alzheimer''s disease is not known for certain.

Immunogen
Anti-human NCSTN mAb is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with recombinant human NCSTN amino acids 34-153 purified from E. coli.

Purification Method
NCSTN antibody was purified from mouse ascitic fluids by protein-G affinity chromatography.

For periods up to 1 month store at 4°C, for longer periods of time, store at -20°C.

Applications
NCSTN antibody has been tested by ELISA and Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:500 ~ 1000. Recommended starting dilution is 1:500.
