Type:
Mouse antibody Monoclonal

Introduction:
The HDV genome exists as a negative sense, single-stranded, closed circular RNA. Because of a nucleotide sequence that is 70% self-complementary, the HDV genome forms a partially double stranded RNA structure that is described as rod-like. With a genome of approximately 1700 nucleotides, It has been proposed that HDV may have originated from a class of plant viruses called viroids. Evidence in support of this hypothesis stems from the fact that both HDV and viroids exist as single-stranded, closed circular RNAs that have rod-like structures. Likewise, both HDV and viroids contain RNA sequences that can assume catalytically active structures called ribozymes.

Specificity:

Mouse monoclonal antibody against the amino acid residues 4-10 (SESKRNR) of HDAg (isolate 25).

Antigen used for Immunizations:
Thehepatitis virus delta antigen-coding fragment (isolateTW2576) was inserted into pPinPoint plasmid andthe expressed recombinant was used for mouse immunization. After cell fusion, the hybridomas obtained were screened for antibody reacting withthe delta antigenfused in different format (MBP-25L) and tested negative forother fusion proteins.

Concentration:
HDV Antibody-HP6A1 is supplied at the concentration of 1 µg/µl.

Storage Buffer:
Theammonium sulfate-precipitated ascites fluid is kept in PBS with 1% BSA and then lyophilized.

Reconstitution ecommendations:
Reconstitute in 18MΩ-cm H2O to a concentration of 1.0 mg/ml.
Stock solutions should be apportioned into working aliquots and stored at <-20ºC.
If more dilute solution required, it is suggested that they be made in a buffered solution containing carrier protein such as PBS with 0.1% BSA.

Reconstitutions on use:
1. Immunofluorescence:
Typical working dilution on frozen sections 1:200. 60 minutes primary antibody incubates at 25ºC.
2. Western blotting:
Recommended: working dilution onWestern blotting 1:500. 60 minutes primary antibody incubates at 25ºC.

Storage and Stability:
Store unopened antibody at 4ºC or lower. Under these conditions, there is no significant loss in product performance one year from the purchase date. The reconstituted antibody is stable for at least two months when stored at 4ºC. For long term storage, it is recommended that aliquots of the antibody are frozen at -20ºC (frost-free freezers are not recommended). Repeated freezing and thawing must be avoided. Prepare working dilutions on the day of use.

Usage:
Prospec's products are furnished for LABORATORY RESEARCH USE ONLY. The product may not be used as drugs, agricultural or pesticidal products, food additives or household chemicals.
