Introduction

Bile salt-activated lipase (BAL or CEL) is a glycoprotein secreted from the pancreas into the digestive tract and from the lactating mammary gland into human milk. The milk of mammals, including humans, contains BAL to enable fat absorption in infants. The Human breast milk contains a bile salt activated lipase at very high levels. The physiological role of CEL is in cholesterol and lipid-soluble vitamin ester hydrolysis and absorption. BAL promotes large chylomicron production in the intestine. CEL’s presence in the plasma suggests its interactions with cholesterol and oxidized lipoproteins to modulate the progression of atherosclerosis. In pancreatic tumor cells, CEL is assumed to be sequestrated within the Golgi compartment and is probably not secreted.

Description

The Carboxyl Ester Lipase produced from Human breast milk has a molecular mass of 79.322kDa (calculated without glycosylation) containing 733 amino acid residues.

Source

Human breast milk.

Physical Appearance

Filtered White lyophilized (freeze-dried) powder.

Formulation

CEL protein filtered (0.4µm) and lyophilized in 0.5mg/ml in 0.05M phosphate buffer and 0.075M NaCl pH 7.4.

Solubility

It is recommended to add deionized water to prepare a working stock solution of approximately 0.5 mg/ml and let the lyophilized pellet dissolve completely. CEL is not sterile! Please filter the product by an appropriate sterile filter before using it in the cell culture.

Stability

Store lyophilized protein at -20°C. Aliquot the product after reconstitution to avoid repeated freezing/thawing cycles. Reconstituted protein can be stored at 4°C for a limited period of time; it does not show any change after two weeks at 4°C.
